Mechanisms for soil moisture effects on activity of nitrifying bacteria.

چکیده
Moisture may limit microbial activity in a wide range of environments including salt water, food, wood, biofilms, and soils. Low water availability can inhibit microbial activity by lowering intracellular water potential and thus reducing hydration and activity of enzymes. In solid matrices, low water content may also reduce microbial activity by restricting substrate supply. As pores within solid matrices drain and water films coating surfaces become thinner, diffusion path lengths become more tortuous, and the rate of substrate diffusion to microbial cells declines. We used two independent techniques to evaluate the relative importance of cytoplasmic dehydration versus diffusional limitations in controlling rates of nitrification in soil. Nitrification rates in shaken soil slurries, in which NH(inf4)(sup+) was maintained at high concentrations and osmotic potential was controlled by the addition of K(inf2)SO(inf4), were compared with rates in moist soil incubations, in which substrate supply was controlled by the addition of NH(inf3) gas. Comparison of results from these techniques demonstrated that diffusional limitation of substrate supply and adverse physiologic effects associated with cell dehydration can explain all of the decline in activity of nitrifying bacteria at low soil water content. However, the relative importance of substrate limitation and dehydration changes at different water potentials. For the soil-microbial system we worked with, substrate limitation was the major inhibiting factor when soil water potentials were greater than -0.6 MPa, whereas adverse physiological effects associated with cell dehydration were more inhibiting at water potentials of less than -0.6 MPa.

Soil Environmental Conditions and Microbial Build-Up Mediate the Effect of Plant Diversity on Soil Nitrifying and Denitrifying Enzyme Activities in Temperate Grasslands
Random reductions in plant diversity can affect ecosystem functioning, but it is still unclear which components of plant diversity (species number - namely richness, presence of particular plant functional groups, or particular combinations of these) and associated biotic and abiotic drivers explain the observed relationships, particularly for soil processes. We assembled grassland communities ...
متن کاملResponse of soil microbiota to the addition of 3 , 3 ′ - diaminobenzidine
3,3′-Diaminobenzidine was applied at doses of 5, 10, 25, and 50 mg kg−1 of soil and their effects were evaluated on indigenous soil microbial communities (viable aerobic bacteria, fungal populations, aerobic N2-fixing bacteria, denitrifying, and nitrifying bacteria), and soil enzymatic activities (acid and alkaline phosphatases, arylsulfatase, and dehydrogenase). At doses of 5 or 10 mg kg−1, 3,...
متن کاملControls on nitrification in a water-limited ecosystem: experimental inhibition of ammonia-oxidising bacteria in the Patagonian steppe
We studied controls on nitrification in an undisturbed water-limited ecosystem by inhibiting autotrophic nitrifying bacteria in soils with varying levels of vegetative cover. The activity of nitrifying bacteria was disrupted using nitrapyrin, 2-chloro-6-(trichloromethyl)-pyridine, under field conditions in three microenvironments (underneath shrubs, next to grasses and in bare soil). Ammonia-ox...
متن کاملProfile Changes in the Soil Microbial Community When Desert Becomes Oasis
The conversion of virgin desert into oasis farmland creates two contrasting types of land-cover. During oasis formation with irrigation and fertilizer application, however, the changes in the soil microbial population, which play critical roles in the ecosystem, remain poorly understood. We applied high-throughput pyrosequencing to investigate bacterial and archaeal communities throughout the p...
متن کاملSoil moisture as a factor affecting the microbiological and biochemical activity of soil
The purpose of this research has been to identify relationships between soil moisture and the growth and development of microorganisms, their diversity and the activity of soil enzymes. Four soils with different texture were analysed. Air-dry soils were watered up to the moisture content corresponding to 20, 40 and 60% of the maximum water capacity (MWC) and subsequently were submitted to deter...
